Blood stem cell transplantation in children and adolescents with acute leukaemia

The main objective of the ALL SCTped 2012 FORUM study is to improve and standardise the treatment of children and adolescents with acute lymphoblastic leukaemia (ALL) through blood stem cell transplantation. The main objective of the FORUM (For Omitting Radiation Under Majority Age) study is to replace radiotherapy, previously considered the standard treatment, with a chemotherapy regimen that is equivalent or potentially superior. To this end, chemotherapy-based conditioning regimens were compared with the standard conditioning regimen, which includes radiotherapy, before blood stem cell transplantation. An interim analysis specified in the study protocol showed that radiotherapy was clearly superior to both chemotherapy variants. Randomised assignment to the different conditioning regimens was therefore discontinued in December 2018. The study is now continuing as an observational study. All key treatment elements and methods in this study are well established in transplant medicine. The study also defines the details of donor selection and treatment during and after transplantation.
