CHIP-AML22 Master Protocol

Acute myeloid leukaemia (AML) is a rare form of blood cancer in children and adolescents. Current treatment has already achieved good results. However, it is important to prevent the disease from not responding to treatment or from returning. For this reason, it is important to study in even greater detail which factors determine the success of the treatment. CHIP-AML22 (Master Protocol) is an international clinical study aiming to improve treatment of children and adolescents with newly diagnosed AML. The study follows the standard treatment approach but also includes new elements. Treatment is given according to risk groups. These groups are determined on the basis of the genetic features of the leukaemia cells observed at diagnosis and how the disease responds to the first phases of treatment. Patients are assigned either to randomised studies within the Master Protocol or to additional linked sub-studies, known as “linked trials”. Assignment may depend on whether a patient meets the criteria for a particular study and also on whether study places are available. The aim is to make it possible to provide improved treatment, tailored to the risk group, for each individual patient, with lower toxicity. This should help to increase the chances of a cure and improve the related quality of life. The primary aim of the trial design is to make recruitment as efficient as possible, as newly diagnosed paediatric AML is a relatively rare form of leukaemia in children. The goal is to further improve event-free survival in children and adolescents with newly diagnosed AML through the use of innovative medication and optimised treatment strategies. Ultimately, the findings from this study are intended to shape the new standard of care for this patient group.
